A Rapid Method for Localization of Tissue Structures or Lesions for Electron Microscopy

Abstract
Adjacent thick and thin serial sections of osmium-fixed methachry-late embedded tissues were used to localize a given structure in both light and electron microscopy. For light microscopy 0.5-u thick sections were placed for few seconds in acetone, hydrated, stained for few seconds in 2% toludine blue, rinsed with water, blotted, rinsed with acetone, cleared with toluene and mounted in permount. Total time of the staining procedure should not exceed 1 minute.
