Aminopeptidase from Brevibacterium linens: Activation and Inhibition

Abstract
Eine gereinigteBrevibacterium linens-Aminopeptidase wurde in bezug auf Aktivierbarkeit und Inhibierbarkeit bei Verwendung vonL-α-Leucyl-4-Nitroanilid undL,-Leucyl-L,-Leucin als Substrat, charakterisiert. Das Enzym wurde durch Kobalt aktiviert, soferne eine Vorbebrütung von Enzym mit Metall erfolgte. Als stark inhibitorisch wirksam erwiesen sich Schwermetalle, metallkomplexierende Agentien, Reduktionsmittel, die Modifikation aromatischer Aminosäuren sowie die Gegenwart hydrophober Substanzen und bestimmter Aminosäuren in den Testansätzen. Unter der Annahme, daß dieser B.linens-Aminopeptidase eine Bedeutung während der Oberflächenreifung von Käsen zukommt, werden die möglichen Auswirkungen spezifisch technologischer Verhältnisse auf deren Aktivität diskutiert. Activation and inhibition of a purified aminopeptidase fromBrevibacterium linens was investigated usingL-α-leucyl-4-nitroanilide andL-leucyl-L-leucine as substrates. The enzyme was activated by cobalt, provided that the enzyme was preincubated with the metal. Strong inhibitory effects were derived from heavy metals, metal-complexing compounds, reducing agents, the modification of aromatic amino acids, and the presence of hydrophobic substances or certain amino acids in the test mixtures. Supposing that thisB. linens aminopeptidase plays a part during surface-ripening of cheeses, possible consequences of specific technological conditions for its activity are discussed.Keywords
