Abstract
Studies were carried out utilizing an isolated saphenous vein in an effort to define the substances active in the "trigger" mechanism of endotoxin shock. Data presented indicate that plasma, platelets, and endotoxin are involved in this mechanism. The plasma factor is dializable, heat labile, and has a molecular weight of from 10,000 to 20,000. The product of this combination resembles histamine in that it is blocked by antihistaminics and its effect on vessel contraction is similar, with the exception of an initial lag period. The fact that platelets are a rich source of histamine adds weight to the thesis that histamine may be the factor involved in the trigger mechanism of endotoxin shock.
