CHROMOTROPE 2R AND PHOSPHOMOLYBDIC ACID AS DIFFERENTIAL STAINS FOR CONNECTIVE AND MUSCULAR TISSUES

Abstract

A new staining method with a color characteristic very near the results obtained with the classical Mallory technique is described. The method can be used after the most commonly employed fixatives. The staining technique is as follows: Deparaffinated and hydrated sections are stained for 10 minutes in a 1% solution of Chromotrope 2R in 1% Acetic acid. They are then rinsed in 3 changes of distilled water, transferred for 1-2 minutes to a 1% solution of Phosphomolybdic acid in distilled water and after that rinsed again in 3 changes of distilled water. The sections are finally transferred to a 1% solution of Stannous chloride in 1/10 N Hydrochloric acid for 1/2-1 minute. They are then rinsed for the last time in 3 changes of distilled water, passed through 3 changes of absolute ethylalcohol, 2 changes of xylol and mounted in the usual way.
