Über den Nachweis von Pregnandiolglukuronid im Harn normaler Frauen und Männer

Abstract
The 24 hr. urine specimens of normal [female] [female] and [male] [male] were thoroughly extracted with butanol, the butanol extract was dried in vacuo on the hot water bath, the residue was dissolved in 20 ml. N/10 NaOH and the soln. extracted 4 times with a total of 20 ml. butanol. This butanol extract was washed with water, dried in vacuo and the residue was dissolved in 3.5 ml. water and 6 ml. acetone. The Na pregnanediol glucuronide was precipitated by adding 60 ml. acetone, the precipitate was dissolved in a little 96% alcohol, the mixture filtered, and the filtrate in a tarred flask dried on the water bath. The product was weighed and the m. p. (260[degree]-270[degree], with decomposition) detd. under the microscope. The Ba salt of pregnanediol glucuronic acid was prepared by dissolving 50 mg. of the Na salt in 100 ml. 50% alcohol and adding 5 ml. of a 5% soln, of Ba acetate in 50% alcohol. The precipitate was recrystallized from 50% alcohol and the Ba salt of pregnanediol glucuronic acid (C27H43O8Ba), m. p. 272[degree], obtained. It was possible to prepare the Ba salt from as little as 1 [gamma] of Na pregnanediol glucuronate and to det. the m. p. of the beautiful crystalline product under the microscope. The solubility of the Na salt in 50% alcohol was 10 times that of the Ba salt, the solubility of the Ba salt being only 28.3 mg. %. The Ba salt of pregnanediol glucuronic acid was prepared from normal urine. In the 2d half of the menstrual cyclej due to the activity of the corpus luteum, normal [female] [female] excreted about 8 mg. of pregnanediol glucuronic acid/day calculated as the Ba salt, while [male] [male] excreted about 0.7 mg. of the compound/1. of urine. In [male] [male], pregnanediol may have its origin in the adrenal cortex, perhaps from desoxycorticosterone or progesterone.This publication has 3 references indexed in Scilit:
