Isolation and Purification of Hepatitis-associated Antigen by Affinity Chromatography with Baboon Antiserum

Abstract
A simple method for the isolation, purification, and concentration of hepatitis-associated antigen (HAA) is described. The technique is based on affinity chromatography in which baboon antiserum is used to trap HAA and 5 m sodium iodide is used to release the antigen. The method was superior to density-gradient centrifugation and ultracentrifugation. Suspensions of HAA with no detectable serum protein and titers of 1:2,000 as determined by immunoelectrophoresis were obtained.
