Abstract
The reaction involves heating the steroids in chloroform with a reagent (containing 0.1 ml 70% perchloric acid, 3 ml absolute alcohol and 7 ml chloroform) and salicylic acid, for 5 minutes on a boiling water bath. The residue is dissolved in a phenol-chloroform mixture, and aliquots added to ether with and without peroxide. The former gives the blank, the latter the experimental value. A calibration curve for 0.40 ug is presented. The extraction of the steroids from urine is described. The recovery of steroids added to urine is only 50%.
