A method for the determination of nucleotide sequence in polyribonucleotides

Abstract
The terminal nucleotide of the polyribonucleotide is dephosphorylated with phosphomonoesterase. The exposed adjacent hydroxyl groups are oxidized with periodate and incubation at pH 10 splits off the oxidized nucleoside residue. By repetition of this process nucleotides can be removed singly until only one remains. The oxidized nucleosides are identified by paper chromatography. The method was applied successfully to 5 dinucleotides and 3 trinucleotides.Keywords
