DNA in Grasshopper Spermatogenesis, Oögenesis, and Cleavage

Abstract
Amounts of desoxyribose nucleic acid (DNA) in individual nuclei of the grasshopper Melanoplus differentialis were detd. photometrically using the Feulgen reaction. In all cells studied there was an approx. constant relationship between the number of chromosomes present (haploid or diploid) and the amt. of DNA. In dividing tissues (gonia and embryonic cells) DNA was apparently doubled in interphase preceding mitosis. Results thus support the concept that DNA is an essential gene component.
