The Preparation of Human Kininogen and the Elicitation of Antibody for Use in a Radial Immuno-Diffusion Assay

Abstract
Kininogen was purified from fresh human plasma using mild conditions and an inhibitor of the kinin-activating sequence. The material recovered after chromatography, gel filtration and electrophoresis was only of the low molecular weight variety. It has been shown to release kinin upon treatment with purified human plasma kallikrein, urinary kallikrein or trypsin. Antibody elicited in rabbits with purified kininogen reacted with biologically active kininogen eluted from disc gel fractions. This antibody was employed in the preparation of a radial immunodiffusion assay for kininogen in human biologic fluids.
