A Paper Chromatographic Method for the Detection of 3-Amino-1,2,4-Triazole in Plant Tissues.

Abstract
Extracts of leaves from Black Valentine bean and Baart wheat plants previously treated with 3-amino-l,2,4-triazole (ATA) were made with 80% ethanol, spotted on filter paper, and developed in an ascending system with a solvent mixture of ethanol: water: 1-butanol, 1:1:4 (v/v). After drying, the chromatograms were sprayed with: (1) a 5% (w/v) aqueous solution of KNO2, then (2) a saturated solution of purified phenol in 20% (v/v) HC1. Yellow spots containing free ATA appeared at Rf 0.45 - 0.48. Bound ATA appeared at Rf 0.025 - 0.028. Sensitivity of the method was found to be 0.1 [mu]g of ATA per spot.
