Response of PAH Transport and Oxygen Consumption in the Rabbit Kidney Slice to Graded Doses of X-Ray and Several Metabolic Inhibitors

Abstract
The ability of rabbit kidney cortex slices to develop a concentration gradient of para-aminohippuric acid (PAH) was used as a measure of the effects of X-rays, DNP, HgCl2 and carinamide on the transport mechanism. The responses to graded doses of these agents are sigmoid, and can be resolved effectively by probit analysis using the log dose. Estimates of the ED50 appear to be independent of many of the variables that affect the absolute concentration ratio. X-rays in doses over 25,000 r were found to cause a progressive increase in O2 consumption of the slice in a manner similar to but not identical with DNP. The significance of these findings was discussed from the viewpoint that this X-ray response represents acute cytoplasmic damage of the renal tubular cell.Keywords
