An enzyme hydrolyzing thiamine diphosphate (cocarboxylase) to thiamine monophosphate has been partly purified from rat liver. The pH optimum is 9.3, and it requires the presence of Mg or Mn ions. Of inhibitors tested only fluoride strongly inhibits enzyme ac-tivity.
