Abstract
The crude coenzyme obtained from pig heart muscle by the method previously described (Banga, Gyorgyi & Vargha, ibid. 210: 228) amounted to about 800 gm. crude product containing 4.75% P from 5 kgm. muscle. This was dissolved in 50 times its wt. of 1% HC1, 1 gm. of phos-photungstic acid added and the ppt. repeatedly extracted with 20 cc. acetone followed by HCl-acetone-ether, and drying. Yield 25%, P 4-4.7%. The powder was dissolved in the least possible amt. of water, centrifuged, neutralized with NaHCO3, an equal wt. of picric acid in hot conc. CH3OH soln. added, the mixture dried in a vacuum desiccator and the residue extracted with acetone to remove excess picric acid. The residue was again dissolved in hot water, centrifuged hot and cooled yielding the coenzyme picrate. Yield 50%, activity 6 times that of crude product. The coenzyme was a monophosphate of a monadenylnucleotide which was not identical with muscle adenylic acid or adenylpyrophosphate, but possibly identical with Euler''s cozymase.This publication has 1 reference indexed in Scilit:
