RESOLUTION OF THE E. COLI AMINO ACYL sRNA TRANSFER FACTOR INTO TWO COMPLEMENTARY FRACTIONS

Abstract
The separation of two complementary factors (A and B) for phenylalanine polymerization from phenylalanyl sRNA on E. coli ribosomes is described. The supernatant of Alumina-ground E. coli paste, after preliminary purification yields, on elution from DEAE-Sephadex, factor A with 0.2 M phosphate and factor B by raising phosphate concentration to 0.3 M. Further purification of B is difficult because of instability, whereas factor A is more stable. A lag in onset of polymerization was frequent in the reconstituted system.Keywords
