Trimethoprim-Sulfamethoxazole: Absorption, Excretion, and Toxicity in Six Children

Abstract
Five infants, seven to 17 months old, and an 11-year-old child were treated with 185 mg of trimethoprim (TMP) plus 925 mg of sulfamethoxazole (SMZ) per m2 daily in two equal doses. Peak levels were observed in the blood between 1 and 4 hr, and high levels of activity were sustained for 12 hr after the first and subsequent doses. There was no significant accumulation of TMP and SMZ. A beneficial therapeutic effect was achieved in all six patients. Granulocytopenia was observed in one child, the recipient of a renal transplant who was receiving immunosuppressive drugs concurrently.
