Abstract
Rapid initial absorption followed by slow, linear absorption of hexestrol from compressed tablets is observed following subcut. implantation or in vitro incubation with water or plasma. This effect, seen best with 1000-250 mg. tablets, is obscured or absent in 50-15 mg. tablets. Early in the period of contact with plasma the tablets become infiltrated with insoluble protein ghosts which are distinct from connective tissue capsules. On applying corrections for the wt. of the ghost, the curves for small tablets in rats are of the same form as for larger tablets. The absorption rate during the linear phase is characteristic for the given wt. class of tablet. Absolute rates/mm.2 for various sizes in different animals are approx. constant. Large tablets in small animals, however, show very reduced or nil linear absorption, possibly due to saturation of body with hexestrol during initial phase.
