Estimation, Isolation and Identification of Auxins in Plant Material

Abstract
The procedure was as follows: Dry whole wheat kernels are soaked for 4 hrs. in NaOH solution at pH 10.5, ground in a glass mortar with clean sand, allowed to stand at 20[degree] C for 45 hrs. (together with 1 ml of toluene) with the pH maintained at 10.5. The mixture was then centrifuged and the supernatant liquid tested by the standard Avena procedure. Indole-3-acetic acid and crystalline pseudo-auxin-a were thus isolated from corn.
