Animal and Human Tolerance of High-Dose Intramuscular Therapy with Spectinomycin

Abstract
Spectinomycin produced no evidence of ototoxicity or serious side effects when given im to 15 men at a dosage of 8 g per day for 21 days in a double-blind comparative study. Repeated evaluations of local tolerance showed that the antibiotic caused more pain and induration than im placebo injections of saline. Less discomfort was encountered during the first week of therapy, but most subjects were able to finish the course of treatment without severe discomfort.
