BLOOD ACTH IN THE STRESSED ADRENALECTOMIZED RAT AFTER INTRAVENOUS INJECTION OF HYDROCORTISONE1

Abstract
The concentration of ACTH in blood from ether-anesthetized adrenalectomized rats was determined prior to, and 1/4, 1-1/4, and 4-1/4 minutes after intravenous injection of 1.0 mg hydrocortisone/100 g rat. The blood ACTH titer of the pre-steroid (control), the 1/4 minute and the 1-1/4 minute samples were 9.6 [plus or minus] 1.8, 8.5 [plus or minus] 1.8, and 10.5 - 2.4 m[mu]/ 100 ml; the titer of the 4-1/4 minute sample was 1.6 [plus or minus] 0.4 m[mu]/ 100 ml. In vitro studies indicate that the oxycellulose technique quantitatively recovers ACTH from blood containing 0.08 mg hydrocortisone/ml. It is concluded that (1) intravenously given hydrocortisone can effectively block ACTH release in the stressed adrenalectomized rat within 1-2 minutes, and (2) a high concentration of hydrocortisone in blood will not interfere with the recovery of ACTH.
