Abstract
Using root tips of Narcissus poeticus, salicylic acid and 31/3% phosphoric acid were tried in 20 combinations with 4% formaldehyde, 2.5% chromic acid, and copper hydroxide. 4% phosphoric acid in 4% formaldehyde gave a fixation showing details of chromosome structure. This fixation followed by crystal-violet-iodine appears to be as specific for the chromonema as the Feulgen stain is for chromatin. Chromic acid (0.5-2.5%) in 100 cc. of saturated salicylic acid solution is suggested as a possible histological fixative.
