Endogenously Released Adenosine Regulates Excitability in the In Vitro Hippocampus

Abstract
SUMMARY The role of adenosine in regulating epileptiform discharge was studied in the in vitro hippocampal slice preparation. Exogenously applied adenosine appeared to have anticonvulsant properties (EC50∼ 10 μM), in that it slowed or suppressed spontaneous interictal discharges. In addition, the release of adenosine (or related adenine nucleotides) from the slice preparation appeared to exert a tonic inhibitory influence on these types of discharges. This provides further evidence supporting a role for adenosine as an endogenous regulator of the excitability of the central nervous system, particularly in convulsive states. ZUSAMMENFASSUNG Die Rolle des Adenosins in der Regulierung epileptiformer Entladungen wurde im in vitro-Versuch an Hippokampusschnittpräparaten untersucht. Exogen appliziertes Adenosin schien zwei antikonvulsive Eigenschaften zu haben (EC50∼ 10 μM): Es verlangsamte oder unterdrückte die spontanen interiktalen Entladungen; zusätzlich schien die Freisetzung des Adenosins (oder verwandte Adeninnukleotide) aus dem Schnittpräparat einen tonischen inhibitorischen Einfluß auf diese Art Entladungen auszuüben. Diese Beobachtung bringt einen weiteren Hinweis dafür, daß Adenosin eine Rolle als endogener Regulator der Reizbarkeit des ZNS besonders bei Krampfzuständen spielt.Keywords
