Addiction Potentialities of 1, 1-Diphenyl-1-( -Dimethylaminopropyl)-Butanone-2 Hydrochloride (Amidone) in the Monkey.

Abstract
Chronic daily subcut. injn. of amidone in rhesus monkeys for 75-95 days with maximum doses of 11-13 mg./kg. did not lead to a condition of physical dependence (addiction) such as occurs with morphine. Increased susceptibility to the depressant action of the compd. was noted as the daily injns. progressed. Observations indicated an absence of crossed tolerance from amidone to morphine but did show minimal crossed tolerance from morphine to amidone. In the monkey, amidone produces an acute depression which is maximum in 90 min. and 4-5 hrs. in duration. The lethal dose lies between 10 and 20 mg./kg. and death results from respiratory failure.
