Abstract
Wistar系雄ラットに3'-Me-DABを短期間(1, 2, 4週)投与し,その後普通食で飼育しその肝に起こる変化を経時的に観察した.肝細胞の変化は可逆的で投与中止後回復に向かう.一方異常増殖したoval cellは,大部分萎縮消失するが,一部は結合織の増殖を伴いcholangiofibrosisの結節を形成する.3'-Me-DAB投与中止後も時間の経過に伴い結節はその数・大きさを増す.管腔周囲の線維化により上皮細胞は萎縮し瘢痕化へと進むが,周辺に新結節の形成,胆管の腺腫様増生が起こる.瘢痕化しつつある結節内に嚢胞形成が見られ,上皮細胞に異型的増殖像が散見された.投与中止後40週経った一例に転移を伴ったcholangiocarcinomaの発生が認められた.以上よりcholangiofibrosisの形成にoval cellの増殖が必要であり,cholangiofibrosisは不可逆性増殖を続ける事,その上皮細胞からcholangiocarcinomaの発生の可能性について考擦する.
