The Ribonuclease of Phaseolus Aureus Roxb:I. Purification of the Enzyme

Abstract
The ribonuclease of Phas. aureus Roxb. was purified according to a scheme which involves acid treatment, (NH42SO4 precipitation, adsorption on Ca3PO42 and subsequent differential elution by KH2PO4, selective heat denaturation and chromatography on Amberlite XE-64. Concomitant phosphodiesterases and phosphomono-esterases (non specific phosphatase and 3′-nucleotidase) are eliminated. A purification of about 200 times was obtained in certain chromatographic fractions. Chromatography by pH-variation resulted into a resolution of the RNase activity into three different enzymic fractions.
