Abstract
The lipolytic and antilipolytic effects of dihydroergotamine (DHE) were studied on isolated rat epididymal adipose tissue cells in terms of glycerol and free fatty acid release. The lipolytic effect of DHE was augmented by the addition of theophylline and was suppressed by propranolol, Kö 592, phentolamine, and insulin. DHE blocked the lipolytic effect of adrenaline, but had no effect on lipolysis caused by ACTH, TSH and glucagon. The results suggest that DHE has multiple sites of action at points preceding the formation of cyclic AMP. They appear to be closely related to catecholamine receptor sites and unrelated to receptor sites of other lipolytic hormones. 1 Supported by the Atkinson Charitable Foundation of Toronto, and Hoechst Pharmaceuticals of Canada.
