Therapy of Uncomplicated Gonococcal Urethritis in the Male with a Single Dose of Amoxicillin

Abstract
Amoxicillin was used to treat 98 males with uncomplicated urethritis caused by Neisseria gonorrhoeae. Disease was proven by culture in all patients. A single 1.0-g dose resulted in a 27% failure rate; a 1.5-g dose produced only a 75% cure rate. A 3.0-g dose was administered to 67 of the 98 patients, and the cure rate was 97%, with only two failures. This compares with a cure rate of 93% achieved with 2.4 × 106 units of procaine penicillin. The minimal inhibitory concentrations of amoxicillin and penicillin G were determined for the isolates. The minimal inhibitory concentration of penicillin was slightly lower. Amoxicillin was well tolerated without adverse side effects.
