Abstract
Guanine is deaminized by purified guanase (from rabbit liver), while adenine does not yield NH3. Tissue enzymes are killed by freezing with liquid air. After hydrolysis with 2% H2SO4 and alkalinization with 33% NaOH the NH3 formed is removed by vacuum distillation. When the last traces of NH3 have been removed, the enzyme soln. is added and the whole kept at 40[degree] C for 3 hrs. at pH 8.8. NH3 formed is then removed in vacuo. The guanine content of rabbit liver, muscle and kidney varies from 0.023 to 0.114%, pig spleen contains 0.165% and calves[image thymus from 0.446 to 0.893%.
