Abstract
Slices of liver and kidney 0.2-0.4 mm. thick were prepared from guinea pigs previously fasted for 24 hrs. They were incubated at 38[degree] for 6 hrs. in a reaction mixture (usually 5 ml.) prepared according to Krebs and Henseleit and containing a 0.0025 M conc. of benzoic acid and a 0.1 M conc. of the amino acid. The reaction mixtures were de-proteinized with 0.5 ml. 10% metaphosphoric acid and extracted with ether. The extract was evaporated and the residue hydrolyzed with 2 ml. conc. HCl. Amino N was detd. according to the method of Folin with the step-photometer, filter S 41, and the amt. of hipprtric.acid calculated. Glutamic acid was detd. by the method of Cohen, glyeine by the nitranilate method of Town. Glycine, as-paragine, dl-serine, 1( + )-glutamic acid, glutamine, glycyl-glycine and hydroxyproline all formed compounds with benzoic acid. One of the compounds formed by glutamine gave glycine when hydrolyzed and was considered to hip-puric acid. Hippuric acid was probably also formed from serine.
