Axenic culture of Laminariales in defined media

Abstract
A method for obtaining axenic cultures of laminarialean meiospores for subsequent culture is described. The method involves surface sterilization of the sorus prior to spore release using an antibiotic mixture (622.5 mg penicillin G, 250.0 mg streptomycin sulphate, 100.0 mg chloramphenicol in 1 liter autoclaved seawater) for 3 hr, followed by a 20–30 minute treatment in 1% NaOCI (v/v autoclaved sea water). Axenic gametophyte cultures were obtained for Laminaria saccharina, Alaria marginata, and Nereocystis luetkeana. The sole attempt to obtain axenic culture of Costaria costata failed.
