Efficacy of 1-Ethanesulfonyl-4-Ethyl-Piperazine Hydrochloride in Treatment of Controlled Hemorrhagic Shock.

Abstract
Healthy mongrel dogs paired as nearly as possible for sex, age, general breed and wt. were anesthetized with 30 mg. of pentobarbital/kg. and acute hypotension was produced by controlled bleeding to approx. 40 mm. Hg. The piperazine compound was admd. in doses of 50-100 mg./kg. given in 10-15 min. within the 1st 30 min. or after 90 min. of hypotension. In 1 type of expt. total survival time was found in the control animals to avg. 157 min. and in the exptl. (drug-treated) dogs 240 min. with early treatment, but with no significant difference when treatment was postponed until 90 min. In a 2d type of expt. recovery of the animals was sought by re-infusion of the withdrawn blood after 90-180 min. of hypotension, treatment with the drug having been made as previously. Again highly significant statistical data was obtained with early treatment with the piperazine but none when it was withheld until 90 min. Three dogs of 30 used in the tests were considered to have died from toxic effects of the piperazine.
