Abstract
The intensely fluorescent DANS [1-dimethyl-amino-naphthalene-5-sulfonic acid] derivatives of spermine and spermidine were prepared by the reaction of the polyamines with 1-dimethyl-amino-naphthalene-5-sulfonyl chloride. After the reaction of a tissue homog-enate or extract with DANS-C1, the DANS-derivatives of the polyamines can be separated from all the other fluorescent components, which are present in comparable concentrations, by 1 dimensional thin-layer chromatography on Kieselgel. The DANS-derivatives were determined quantitatively by elution from the Kieselgel layer, or by direct measurement of the fluorescence intensity on the plates. If the thin-layer plate is evaluated directly, as little as 5-10-12 mole of polyamine per spot can be measured with a reproducibility of [plus or minus] 4-6%.This publication has 10 references indexed in Scilit:
