Abstract
The effect of chloramphenicol (CAP) on the assembly of ribosomes into polysomes and on the functioning of these polysomes has been examined in growing cultures of Escherichia coli. The following conclusions are made: CAP does not cause the disappearance of polysomes or prevent the assembly of ribosomes into polysomes. It therefore does not inhibit the binding of ribosomes to messenger RNA. Polysomes formed in the presence of CAP do not form nascent protein. Thus, the antibiotic does not act simply by blocking the release from the rlbosome of finished proteins. The growth of nascent polypeptides is inhibited by CAP, indicating that the antibiotic in some way interferes with a step common to the formation of all peptide bonds of a protein.
