Standardized Paper Electrophoresis of Parotid Saliva

Abstract
A study of the effects of technique factors: dialysis, sample application, voltage-current conditions and staining procedure, was made to establish a routine procedure for paper electrophoresis of human parotid saliva in Veronal buffer, pH 8.6, ionic strength 0.05. The optimum conditions which will give reproducible resolution for the same sample were 300V, 4-4.5 ma (constant voltage) for 17 hours in a Spinco Model R Unit. The strips were stained with Nigrosine. A study of 50 subjects showed that a flow rate greater than 1 ml/min for a single parotid gland was required for adequate electrophoretic resolution. It appears that the active functioning of the gland is reflected in the number of components observable under standardized conditions of analysis, and are characteristic for the individual.Keywords
