Cloning of sporulation gene, spoOF, in Bacillus subtilis with .RHO.11 phage vector.

Abstract
A specialized transducing phage .rho.spoOF, containing one of the stage O sporulation genes, spoOF, of B. subtilis was constructed. Sporulation of spoOF mutant of B. subtilis was restored by lysogenization with .rho.11spoOF, indicating that the mutant allele was recessive to the wild type. The physical map of .rho.11spoOF genome was constructed by using restriction endonucleases BamHI and SalI and compared with that of pll wt genome. The spoOF gene resides on the SalI-B fragment of .rho.11spoOF genome. The 1.3 .times. 106 dalton EcoRI fragment of .rho.11spoOF DNA contained the spoOF gene.
