ISOLATION OF HYALURONATEPROTEIN FROM HUMAN SYNOVIAL FLUID*

Abstract
Hyaluronate was isolated from normal synovial fluid by passing the fluid through a column containing hydroxylapatite and IRC-50 resin, and ultrafiltering the eluate on millipore filters (0.1 [image] pore diameter). The isolated hyaluronate consistently contained 2% protein, and the product was called hyaluronateprotein. Their film combination was shown as follows the product was labeled with Iodine-131, dialyzed to remove free iodine, and subjected to zone electro-phoresis on blocks of Geon resin (427). Over a pH range of 11.2 to 3.5 hyaluronate (measured as hexuronic acid) and protein (measured by radioactivity) migrated as a single peak toward the anode. Neither high salt concentration, urea, nor addition of a basic protein dissociated hyaluronate and protein.This publication has 24 references indexed in Scilit:
