Vereinfachtes Verfahren zur Isolierung von Neuraminsäure als Methoxy-neuraminsäure mit Hilfe von Ionenaustauschern

Abstract
Bovine submaxillary mucin is auto-claved (105-110[degree]C) for 3 hours with 5% methanolic HC1 and the methoxy-neuraminic acid ester then saponified with 2N ammonia. Chloride ions are removed by passage through a column of Dowex 2-X10 (hydrbxyl form), and NH3 in the vacuum desiccator over H2SO4. The product is then applied to a column of Dowex-50-X4 (H+ form, 20-50 mesh), and elution carried out with water. The fractions containing methoxy-neuraminic acid are collected, concentrated and precipitated by addition of ethanol. The product is decolorized by passage through an alumina column and is recrystallized; 360 g of mucin yielded 23 g of methoxy-neuraminic acid.
