Studies on sulphatases. 6. The localization of arylsulphatase in the rat-liver cell

Abstract
Rat liver was homogenized and fractionated by centrifugation in hypertonic and hypotonic sucrose, acetate-buffer, and isotonic KCl. Arylsulfatase activity was found only in the granular material of the cytoplasm (microsomes), none in nuclei or mitochondria. Agglutination of granules at pH 5 resulted in transfer of activity from supernatant to residue. The synthesis of the sulfate of p-hydroxyacetophenone was demonstrated in supernatants of liver extracts after centrifuging at 18,000 g to remove all but a trace of sulfatase activity.
