Centrifugal Sephadex Procedure for Fractionation of Concentrated Skimmilk, Whey, and Similar Biological Systems

Abstract
A centrifugal Sephadex procedure was employed to separate concentrated skim milk and whey systems into 2 fractions on the basis of molecular weight. Centrifugal force (1,000 x g) was applied to a bed of Sephadex G-25 in a basket centrifuge to spin out the colloidal portein-salt components of the sample which were excluded from the interior volume of the gel and the soluble compnents were then eluted from the gel at slow speed (60 x g) with water. The procedure permits fractionation of relatively large volumes (V < 1/2 bed volume) of viscous systems, such as 30-40% concentrated skim milk or whey, in a total time per cycle of N30 min. and with a minimal dilution of the colloidal fraction.
