HPLC-Methode zur Bestimmung von vitamin C in Milch, Molke und Molkegetr nken

Abstract

A method is described for the estimation ofl-ascorbic acid andl-dehydroascorbic acid in milk, whey, and whey drinks by paired-ion reversed-phase HPLC using a C₁₈-column.l-ascorbic acid is detected UV-spectrophotometrically at 248 nm,l-dehydroascorbic acid is determined spectrofluorometrically with excitation at 350 nm and emission at 430 nm, after derivatisation into a quinoxaline fluorophor. Es wird eine lonenpaar-HPLC-Methode zur Bestimmung vonl-Ascorbinsäure undl-Dehydroascorbinsäure in Milch, Molke und Molkegetränken beschrieben. Nach der Trennung an einer C₁₈-Umkehrphasensäule erfolgt der Nachweis vonl-Ascorbinsäure mittels UV-Detektors bei 248 nm,l-Dehydroascorbinsäure wird als Chinoxalinderivat fluorimetrisch bei 350 nm Anregungs- und 430 nm Emissionswellenlänge erfaßt.
