THE HYDROLYSIS OF THE CONJUGATED OESTROGENS IN HUMAN URINE

Abstract
Extraction of estrogens from urine is increased by hydrolysis of the estrogen conjugates. The most effective hydrolysis appears to occur with 15 vol. % HC1, boiling for 60 min. Destruction of estrogens is reduced by the addition of a reducing substance (e.g., Zn, ascorbic acid) or by protection from oxygen by autoclaving in a steam atmosphere, or boiling under N2. Enzymes (glucuronidase or sulfatase) may be used, but since the type of conjugate appearing in the urine varies, enzymatic hydrolysis may be unreliable The authors believe that the increased estrogens extractable after 60 min., 15% HCl hydrolysis as compared with 10 min. hydrolysis (Tzn/To of Smith and Smith), is without physiol. significance, and simply represents improved hydrolysis and extraction.Keywords
